Quantitative PCR issues


Most assays, including qPCR, aren’t meant to determine if something is absent, or negative - the user has to determine some threshold for level of detection, and below that is really “not detectable”, which doesn’t always mean negative. It doesn’t surprise me researchers have struggled with where to set the thresholds - it’s a balance between max sensitivity and reducing false positives
